    Biomedical applications of metal oxide nanoparticles in aging and age‑associated diseases

    Metal oxide nanoparticles are known to exhibit unique properties such as catalyzing the neutralization of superoxide anions, hydroxyl radicals, hydrogen peroxides and behave as antioxidant enzymes. Oxidative stress, damage and chronic inflam mation are major causes and consequences of aging and age-associated disorders. With the increasing popularity of metal oxide nanoparticles, they have been applied in various age-related pathologies using their antioxidant property. Metal oxide nanoparticles have been used as diagnostic, therapeutic, and as theranostics. This review summarizes the applications of metal oxide nanoparticles in aging and age-associated disorders such as cardiovascular diseases, diabetes, cancer, neurodegenera tive disorders. Oxidative stress plays a central role in the activation of inflammatory pathways, disturbing the mitochondrial function, decreasing the telomere length and leading the cell towards senescence or death. Oxidative damage is the common pathway in the progression of aging and related diseases. Metal oxide nanoparticles scavenge or precisely detect the gener ated reactive oxygen species, hence applied in both diagnostics and therapeutics.

    Curcumin ameliorates oxidative stress in red blood cells during ageing

    An essential dietary flavonoid known as curcumin has positive health effects and inhibits the synthesis of reactive oxygen species (ROS). The aim of this study is to look at signs of oxidative stress in red blood cells (RBCs) treated with curcumin as a function of age. A total of 116 healthy volunteers ranging in age from 20 to 81 years provided clinically pertinent blood samples for the investigation. Three groups of subjects were created: young (20 to 35 years), middle (36 to 60 years), and old (>60 years). Oxidative stress was induced in vitro by incubating RBCs with 10-5M tert-butyl hydroperoxide (t-BHP). Malondialdehyde and reduced glutathione were detected by co-incubating RBCs with curcumin (10-8 M to 10-5 M final concentration) and t-BHP to assess the effect of curcumin. After being incubated with t-BHP, the results revealed higher MDA levels (P <0.001) in comparison to their respective controls and the GSH level significantly (P <0.001) decreased during ageing. By raising GSH and lowering MDA levels, curcumin treatment in vitro considerably (P <0.01) mitigated the harmful effect of oxidative stress in RBCs from all age groups. The results of this study showed the potential role of curcumin in the ageing process and it will facilitate the quick screening of novel chemical compounds that may protect RBCs from oxidative stress.

    Ellagic acid: insight into its protective effects in age‑associated disorder

    The disparity in the free radical generation and the production of antioxidants to counteract its effect is known as oxidative stress. Oxidative stress causes damage to the macromolecules such as lipids, carbohydrates, proteins, and DNA and RNA. The oxidative damage to the cellular components leads to a process of aging and various age-associated disorders. The lit erature survey for this review was done using PubMed, Google Scholar, and Science Direct. The papers showing the studies related to aging and age-associated disorders have been selected for reviewing this paper. Ellagic acid has been used as the keyword, and more emphasis has been put on papers from the last 10 years. However, some papers with significant studies prior to 10 years have also been considered. Almost 250 papers have been studied for reviewing this paper, and about 135 papers have been cited. Ellagic acid (EA) is present in high quantities in pomegranate and various types of berries. It is known to possess the antioxidant potential and protects from the harmful effects of free radicals. Various studies have shown its effect to protect cardiovascular, neurodegenerative, cancer, and diabetes. The present review focuses on the protective effect of ellagic acid in age-associated disorders. The effect of EA has been studied in various chronic disorders but the scope of this review is limited to cancer, diabetes, cardiovascular and neurodegenerative disorders. All the disease aspects have not been addressed in this particular review.

    Endometriosis and Depression: A Double Agony for Women

    Abstract Background: Endometriosis is defined as a condition in which a formation of abnormal endometrial tissue outside the uterus takes place. Irrespective of any ethnic and socioeconomic class, the prevalence of the diseases has been seen among women of reproductive age. Endometriosis has been seen adversely affect the physical, psychological, social, and career of women. Summary: This paper aims to identify and describe the experiences and outcomes of endometriosis healthcare by reviewing the existing literature on social and psychological effects of endometriosis. The study serves the purpose of providing insights on women’s dual suffering (mental and social health) and critical comments on the present state of knowledge in order to make future recommendations for psycho-social research. The review included systematic search of the articles from various disciplines like, biology, psychology, sociology, anthropology, etc. A structured process of screening with specific inclusion and exclusion criteria was used to select the articles. The analysis of the articles resulted that woman diagnosed with endometriosis reported poor quality of life and the following symptoms such as anxiety, stress, Chronic Pelvic Pain (CPP), anxiety, dyspareunia, and dysmenorrhea. However, depression appears prominent among women diagnosed with endometriosis. There are few strategies mentioned that can be used to deal with endometriosis. Key Message: The most promising causes of endometriosis include abnormality in immune functioning, atypical endometriotic growth, genetics, epigenetic, embryogenetic theory, and endocrine disruptors. The ill effects have been noted in the following domains of women’s life such as work, close relationships, social well-being, and education, deteriorating their quality-of-life manifold. Psychological intervention is required to deal with the disorder as only medical treatment with pain may not be sufficient. Though, the condition can be improved by providing awareness regarding the severity of the disorder at the school and community levels.

    Oncogenes and tumor suppressor genes: functions and roles in cancers

    Cancer, being the most formidable ailment, has had a profound impact on the human health. The disease is primarily associated with genetic mutations that impact oncogenes and tumor suppressor genes (TSGs). Recently, growing evi dence have shown that X-linked TSGs have specific role in cancer progression and metastasis as well. Interestingly, our genome harbors around substantial portion of genes that function as tumor suppressors, and the X chromosome alone harbors a considerable number of TSGs. The scenario becomes even more compelling as X-linked TSGs are adaptive to key epigenetic processes such as X chromosome inactivation. Therefore, delineating the new paradigm related to X-linked TSGs, for instance, their crosstalk with autosome and involvement in cancer initiation, progression, and metastasis becomes utmost importance. Considering this, herein, we present a comprehensive discussion of X-linked TSG dysregulation in various cancers as a consequence of genetic variations and epigenetic alterations. In addition, the dynamic role of X-linked TSGs in sex chromosome–autosome crosstalk in cancer genome remodeling is being explored thoroughly. Besides, the functional roles of ncRNAs, role of X-linked TSG in immunomodulation and in gender-based cancer disparities has also been highlighted. Overall, the focal idea of the present article is to recapitulate the findings on X-linked TSG regulation in the cancer landscape and to redefine their role toward improving cancer treatment strategies.

    Protective effect of ellagic acid on erythrocytes subjected to oxidative stress during human ageing

    In the present study, the potential role of ellagic acid on various biomarkers of oxidative stress during human ageing has been studied. The study was carried out on 75 healthy individuals of both sexes. They were divided into three age groups viz., young (<36 years; n=23; male=13, female=10), middle (36-60 years; n=38; male=25, female=13), and old (>60 years; n=14; male=8, female=6). The oxidative stress was induced by using tert-butyl hydroperoxide (t-BHP). In each age group, erythrocytes were incubated with ellagic acid (final concentration 10-5M). The result demonstrated a significant (P <0.01) decrease in total antioxidant potential in terms of FRAP value during ageing. A significant (P <0.05) in vitro protective effect of ellagic acid on malondialdehyde and reduced glutathione levels on erythrocytes against oxidative stress induced by tert-butyl hydroperoxide has been reported. The study was performed on erythrocytes of different age groups. The findings suggest a possible role of ellagic acid as an anti-ageing compound.

    Recent advances in the protective role of metallic nanoparticles in red blood cells

    The interaction of nanoparticles with the biological system has increased with the increasing popularity of nanomedicines. Red blood cells (RBCs) are very sensitive, and abundant cells in the blood. They are highly prone to oxidative damage due to constant interaction with oxygen itself, foreign particles in the blood, and the lack of repair mechanism. The cell mem brane of RBCs undergoes lipid peroxidation, protein oxidation, and heme degradation which results in altered membrane permeability, changes in the morphology, and functioning of RBCs. The nanoparticles induce oxidative stress, hemolysis, morphological changes, membrane deformability, and alterations in hemoglobin structure in RBCs. In this review, the effects of metallic nanoparticles and their modifications on the physiology, and life span of RBCs are discussed. The detailed analy sis of the antioxidant enzymes-like activity of metal nanoparticles is expected to highlight the beneficial use of these metal nanoparticles in RBCs against oxidative stress and the development of new biosafe nanodrugs.

    Sukshmjeevanu in Vedas: The Forgotten Past of Microbiology in Indian Vedic Knowledge

    No one questions the existence of presumptive knowledge of invisible organisms causing disease, decay and destruction mentioned before the discovery of the microbial world by Antonie Van Leeuwenhoek, who was the first to describe the invisible world as per literature available today. However, the knowledge about microbial world by Indian Rishis presented in Sanskrit shlokas or suktas of our traditional manuscripts such as Vedas remained unseen, where the Rishis had predicted the role of microorganisms known as Krimi or Jeevanu years before Leeuwenhoek. This note is an attempt to bring an emphasis to revisit our traditional Vedic knowledge and establish them through research based facts for wider acceptance globally.
